Reproductive Processes Russell, Scott [1], Larson, Preston [2]. Use of cryoEM in assessing developmental mechanisms of rice reproduction. Rice (Oryza sativa L.) is the most widespread crop in the world, and one that is useful for understanding reproductive mechanisms, as it is also a model plant for molecular biology and one for which -omic characteristics of flowering plant gametes are known. In addition to this significance, rice can also be used as a model for enhanced resolution electron microscopy that can be achieved using high resolution imaging of frozen material using cryotomographic equipment and sensitive direct electron imaging in resolutions that approach near atomic resolution under the best conditions. Such imaging may provide more sensitive imaging of dynamic developmental processes than are possible using traditional transmission electron microscopy. Such TEM images in the past are fraught with high damage, chemical artifacts of such fixatives as glutaraldehyde and osmium tetroxide and dehygration that provide significant artifacts. The current study represents an extension of these methods to achieve imaging that more closely records the native condition of tissues, cells and molecules at higher resolution.
