Antibacterial and Antioxidant Activity of Polyphenols Extracted from Four Species of Gymnopilus (Macro-fungi) Collected from the Wild in Nigeria.

Mushrooms are reservoirs of natural compounds with potential medical benefits that are under-utilized. Gymnopilus species occur abundantly in the wild in Nigeria and their uses are not known. The medicinal value of the species of Gymnopilus frequently encountered in the wild in Nigeria was investigated. The inhibitory activities of Gymnopilus species against bacteria and free radicals (DPPH) was evaluated in the current study. Antibacterial activity was assessed with both gram-positive (Staphylococcus aureus and Listeria monocytogenes) and gram-negative (Escherichia coli, Salmonella enterica and Klebsiella pneumoniae) bacteria. The paper disc diffusion method was compared to the microdilution method of evaluating antibacterial activity and the minimum inhibitory concentration (MIC) of the extracts determined. The total phenolic content varied in the different species from 2.88±0.03mg/g GAE in Gymnopilus delipis (Berk & Broome) Singer to 9.25±0.64 mg/g GAE of dry sample in Gymnopilus lepidotus Hesler. The antioxidant activity (inhibition of free DPPH radicals) varied from the highly reactive G. lepidotus (82.77%) to the relatively less reactive -G. suberis (54.64%). The Trolox equivalent capacity of the extracts ranged from 86µg TE/mg dry sample in Gymnopilus purpureosquamulosus Hoil to 210µg TE/mg dry sample in Gymnopilus delipis. The half maximal inhibitory concentration (EC50 values) for all extracts tested was above 5.00mg/ml. The extracts also varied in their antimicrobial potential with the minimum inhibitory concentration ranging from 5.00-15.00 µg/ml. The extracts of G. lepidotus, G. suberis and G. delipis were active with both gram-positive and gram-negative bacteria tested. The extracts of G. purpureosquamulosus Hoil recorded higher activity with gram-negative bacteria. The polyphenol extracts of the four Gymnopilus species tested were comparable to standard antibiotics and are potential effective antioxidant agents.
