Comparative Genomics/Transcriptomics

The first genome sequence for Penstemon (Plantaginaceae).

Penstemon is a genus of >285 species, and is endemic to North America. It has a base chromosome number of n=8, with genomes ranging from diploid to dodecaploid. Most species in the genus are diploid. Subgenus Penstemon sect. Penstemon and subg. Saccanthera sect. Saccanthera have polyploid lineages with chromosome counts up to 12x. Genome size for species in Penstemon range from 462 Mbp (P. dissectus, 2x) to 3152 Mbp (P. digitalis, 12x). Here we report on the first genome sequence for Penstemon. Using a combination of PacBio and Illumina sequencing data, via Dovetail Genomics, we acquired whole genome sequence data for P. petiolatus (729 Mbp, 2x). Penstemon petiolatus is positioned near the middle of the phylogenetic tree for the genus, and we were able to collect a sufficient amount of fresh tissue on liquid nitrogen to enable the high molecular weight DNA extraction required by Dovetail Genomics. The resulting genome sequence data included 4.9M reads and roughly 81.5x coverage. We then used the Maker-P pipeline for annotation and conducted comparative genomic analyses with other species in the order Lamiales to investigate patterns of gene and genome duplication.
