Genetic diversity and the population structure in Viburnum erosum (Adoxaceae).

Viburnum erosum Thunb. (Adoxaceae) is a deciduous shrub which is distributed in southern China, Taiwan, Korea, and western Japan. It is characterized by having scaled buds, a pair of stipules, extrafloral nectaries at the base of the abaxial leaf surface, and the red drupes within Viburnum. Plants of V. erosum have been cultivated as an ornamental and used as a source in traditional medicine. Microsatellite markers were developed to examine the genetic diversity and the level of population differentiation of V. erosum. We screened 226 potential microsatellite loci from next-generation sequence data and selected ten polymorphic markers. As a preliminary investigation, we included 40 samples from two populations in Korea (Jeonnam and Jejudo), 20 from Kyushu in Japan, and 20 from Mt. Jinggangsan in China. A total of 73 alleles with a size range of 81-232 bp were identified. An analysis of molecular variance (AMOVA) showed that the variation within populations was higher (92% of the total variance) than the variation between populations (8%), suggesting that gene flows among populations are limited. A principal coordinates analysis of the alleles showed that the Chinese population is clearly separated from the Korean and Japanese populations by the first axis and that the two Korean populations were weakly separated from the Japanese population by a combination of the first and second axes. Our results suggest that the geographical structure in V. erosum is associated with the mode of the dispersal of seeds by birds.
