Phylogenomics Charboneau, Joseph [1], Cronn, Richard [2], Liston, A. [3], Wojciechowski, Martin [4], Sanderson, Michael [5]. Chloroplast phylogenomics in the New World clade of Astragalus L. (Fabaceae). With over 3,000 described species, Astragalus L. (Fabaceae) is one of the largest genera of seed plants. A clade of about 450 species called Neo-Astragalus make up the majority of Astragalus species in the Americas. Neo-Astragalus is estimated to be only about 4.5 million years old, and although the monophyly of the clade itself is well supported, phylogenetic relationships within Neo-Astragalus have remained largely unknown due to low levels of sequence variation seen in genes sequenced thus far. Using genome skimming data, we assembled the chloroplast genomes of three Neo-Astragalus species: Astragalus agnicidus Barneby, Astragalus flexuosus (Hook.) Douglas ex G. Don, and Astragalus mollissimus Torr. The plastomes of the latter two species contained large inversions, and the presence of these inversions in other species may be useful in delimiting major groups within Neo-Astragalus. We screened for these inversions in other Neo-Astragalus species, and we have also sequenced genomic libraries of over 90 other Neo-Astragalus species, from which we are assembling additional chloroplast genomes.
