Biodiversity Informatics & Herbarium Digitization Rosati, Ethan M [1], Samuel, Sam [1]. Undergraduate Student Learning With Extraction of DNA from Herbarium Specimens. The purpose of this project is to extract and sequence DNA that is collected from plant vouchers that are housed within the herbarium at Utah Valley University herbarium (UVSC). We extracted DNA by two methods. One was by using a relatively new technique by way of rubbing the surface of the specimen, lifting cells off the surface of the plants, we consider this non-destructive, the other method of extraction is by grinding a leaf sample. After material collection DNA extraction kits were used on both. We observed the results to compare whether or not the two sampling methods, using the same extraction kits, produce the optimum yields. We then amplified the DNA via PCR to produce more testable yields. We observed the quality and quantity of the PCR products though gel electrophoresis and nanodrop. We used new ITS region barcode specific primers (Cheng, 2016) to investigate success in amplification of the target DNA sequence. We sequenced the ITS region of the collected DNA samples Geneious software was used to observe DNA quality to determine whether the two methods used produced good DNA yield. Ultimately our goal is to add our sequenced data to the barcoding public body of knowledge to help biologists have intellectual control, conserve and protect the biota of Utah and the world.
