Conservation genetics and taxonomy of the southeastern Coastal Plain endemic water lily Nuphar sagittifolia.

Nuphar sagittifolia (Walter) Pursh (Nymphaeaceae), Cape Fear spatterdock, is a southeastern Atlantic Coastal Plain endemic aquatic macrophyte of conservation concern. The taxonomic limits of N. sagittifolia are unclear due to the presence of populations morphologically intermediate between N. sagittifolia s.s. and the widespread Nuphar advena (Aiton) W.T. Aiton subsp. advena. Conservation needs cannot be assessed without a clear circumscription, as the true range and number of populations remain uncertain. Leaf tissue and morphological data were collected from 30 individuals at each of 22 Nuphar populations across the Coastal Plain of North Carolina, South Carolina, and Virginia, in addition to the type populations of N. advena subsp. advena, N. sagittifolia, and N. advena subsp. ulvacea. A combination of morphological and genotyping-by-sequencing data were used to conduct a taxonomic analysis of Nuphar populations in the putative N. sagittifolia range. Genotyping-by-sequencing results provide insight into the genetic structure, extent of clonality, and diversity within and among strict sense populations of N. sagittifolia. Results of this study will enable informed conservation decisions for this charismatic southeastern endemic species.
