Wild hops in the Arizona Sky Islands: Meiotic chromosomes and the structural variability of the Humulus lupulus species. Hop (Humulus lupulus L.) is an important crop worldwide, known as the main flavoring ingredient in beer. The diversifying brewing industry demands variation in flavors, superior process properties, and sustainable agronomics, which are the focus of advanced molecular breeding efforts in hops. Hop breeders have been limited in their ability to create strains with desirable traits, however, because of the unusual and unpredictable inheritance patterns and associated non-Mendelian genetic marker segregation. Cytogenetic analysis of meiotic chromosome behavior has also revealed conspicuous and prevalent occurrences of non-disomic chromosome complexes in prophase, anaphase bridges at meiosis I and II, and unequal distributions of FISH signals at the tetrad stage. These observations together reflect a highly variable structural genome. There are three native, wild varieties of hops in North America: var. neomexicanus, var. lupuloides, and var. pubescens. Given that cultivated hops show extreme meiotic abnormalities that lead to segregation distortion, we asked if wild populations of hop share these traits. To test this, we chose field sites in the Madrean Sky Islands of Arizona, as these hop populations are thought to be isolated and free of cross pollination by feral hop varieties. We tested novel satellite, tandemly repeated DNA sequences as FISH probes to demonstrate that wild hop also exhibits non-Mendelian inheritance of FISH signals at meiosis I and II, as well as chromosome interaction anomalies throughout meiosis.
