Morphological and Molecular Characterization of Cowpea Accessions from Africa.

Morphological and Molecular characterization are crucial tools in assessing the level of genetic diveristy and variation in plant species. In this study, genotypes of cowpea (Vigna unguiculata (L) Walp.) from ten African countries were collected from International Institute for Tropical Agriculture (IITA). The genotypes were planted in a randomized complete block design with four replicates. Morphological characters of seedlings were assessed following the International Board for Plant and Genetic Resource Cowpea Descriptor list and both quantitative and qualitative characters were evaluated. Results showed that the plant habit was primarily sem-erect with intermediate twining (80%) while others have erect and slightly twining habit (20%). Pigmentation ranges from extreme (n=1), moderate= (n=1), slight = (n=16) to none (n=2). Flowers color is either pink (65%), white (20%) or violet (15%). Inflorescence is raceme. Pod is straight (20%) or curved (80%). Peduncle length ranges from 1.87 to 15.5 cm, pod length is 8.63-18.83 cm while seed is 6.10-9.93mm by 3.35-5.75 mm. Analysis of the morphological data using UPGMA showed a dendogram with four (4) clusters. Genomic DNA was successfully extracted from the seedlings with relative absorbance of 1.87 and concentration of 339-923 ng/ul. All the samples were amplified using six SSR markers (SSR27BY, SSR19BY, SSR10BY, SSR9BY, SSR8BY and SSR21BY) however only three successfully amplified. A total of 7 alleles were observed while the PIC ranges from 0.868 to 0.992. Assessment of relationship based on molecular characters revealed a dendogram with two clusters and one outgroup. This study revealed low genetic diversity among the accessions and a relatively high variation based on morphological characters. Agro-morphological traits (quantitative and qualitative) evaluated in this studies proved valuable for cowpea genetic diversity studies.
