Conference Wide Galbraith, David [1], Villalobos-Menuey, Elizabeth [2]. Flow cytometry of plants: a practical introduction to angiosperm genome size measurement. There is considerable interest in cataloging the diversity of existing angiosperm species, particularly given anthropogenic change. A convenient marker for characterizing species is the somatic nuclear DNA content. Some time ago, I developed a quick and accurate method for measuring plant genome sizes, involving flow cytometric characterization of the fluorescence emission of nuclei stained within homogenates using DNA-specific stains. Typically, such measurements were made using large and expensive instruments located in core facilities and operated by trained specialists. With the advent of next generations of small, inexpensive, and reliable flow cytometers, this technology is becoming very much more accessible to botanists, taxonomists, physiologists, and developmental and ecological scientists. In this hands-on workshop, we will first provide an overview of the technology and its applications, and then demonstrate use of a next generation flow cytometer, the Beckman Coulter CytoFLEX, for plant analyses.
