Phylogenomics

Species level phylogeny for the order Cornales based on molecular data from nuclear gene target enrichment.

Cornales is a monophyletic order of flowering plants containing horticulturally important families of plants including Cornaceae (dogwoods), Hydrangeaceae (hydrangeas) among others. The monophyly of Cornales is strongly supported by morphological and molecular data, whereas species-level relationships within the order have not been fully resolved. Previously, relationships among families within the Cornales have been estimated based on plastid genes, and moreover, relationships within the genus Cornus have been estimated from two loci (matK and ITS). Our objective is to test these phylogenetic inferences using multiple nuclear genes in a coalescence-based analyses. In this study, we are using molecular phylogenomic approaches on a data set of 252 single-copy genes on over 100 Cornales species. Preliminary analyses of these data have produced a species tree that preserves the monophyly of Cornales families but disagrees with some species placements. For most of the estimated species phylogeny we see high support values and a high degree of concordance among individual nuclear gene trees. However, some relationships among closely related species and subspecies are not well supported. We hypothesize that the poor resolution of relationships among these species may be due to rapid speciation and inter-specific gene flow. Our findings show the utility of the targeted gene capture bait set for resolving patterns of diversification across the Cornales.
